Ontogeny of Dopaminergic Neurons

چکیده
DOPAMINE IN THE ADULT CNS The developmental functions mediated by dopaminergic signaling are not currently fully appreciated, however, more is known about the influences of DA in the mature brain. Synthesis of DA involves conversion of the amino acid L-tyrosine into L-dopa by the rate-limiting enzyme tyrosine hydroxylase (TH). Subsequent activity of DOPA decarboxylase results in final conversion to DA. DA is widely distributed in the CNS with important projections into the forebrain. The nigrostriatal tract consists of DA neurons with cell bodies located in the substantia nigra (SN) pars compacta and axonal processes terminating in the dorsal striatum. The striatum is a component of the extrapyramidal motor system and plays an essential role in the coordination of locomotor activity. The mesocorticolimbic pathway is another major forebrain dopaminergic projection. This pathway arises in the midbrain ventral tegmental area (VTA) and provides input to the nucleus accumbens, and some limbic regions including limbic cortical regions (medial prefrontal (mPFC) and anterior cingulate cortex (ACC)). These pathways are those important for mediating behaviors associated with motivation, reward (endogenous systems and drug abuse) and reinforcement, as well as cognitive and executive functions including attention .

Malva sylvestris aqueous extract could ameliorate 6-hydroxydopamine-induced motor asymmetry with no protective effect on dopaminergic nigrostriatal neurons in the rat
Background and Objective: Parkinson’s disease (PD) is a common neurological disorder due to degeneration of dopaminergic neurons within pars compacta of substantia nigra (SNC). With regard to protective effect of Malva sylvestris (MS), this study was conducted to evaluate the effect of aquaeous extract of this plant in an experimental model of PD induced by 6-hydroxydopamine (6-OHDA). Material...
متن کاملBirth, life and death of dopaminergic neurons in the substantia nigra.
Understanding the ontogeny of A9 dopamine (DA) neurons is critical not only to determining basic developmental events that facilitate the emergence of the substantia nigra pars compacta (SNc) but also to the extraction and de novo generation of DA neurons as a potential cell therapy for Parkinson’s disease. Recent research has identified a precise window for DA cell birth (differentiation) in t...
متن کاملComparison of Rat Primary Midbrain Neurons Cultured in DMEM/F12 and Neurobasal Mediums
Introduction: Midbrain dopaminergic neurons are involved in various brain functions, including motor behavior, reinforcement, motivation, learning, and cognition. Primary dopaminergic neurons and also several lines of these cells are extensively used in cell culture studies. Primary dopaminergic neurons prepared from rodents have been cultured in both DMEM/F12 and neurobasal mediums in several ...
متن کاملTransient Dysregulation of Dopamine Signaling in a Developing Drosophila Arousal Circuit Permanently Impairs Behavioral Responsiveness in Adults
The dopamine ontogeny hypothesis for schizophrenia proposes that transient dysregulation of the dopaminergic system during brain development increases the likelihood of this disorder in adulthood. To test this hypothesis in a high-throughput animal model, we have transiently manipulated dopamine signaling in the developing fruit fly Drosophila melanogaster and examined behavioral responsiveness...
متن کاملTransient activation of dopaminergic neurons during development modulates visual responsiveness, locomotion and brain activity in a dopamine ontogeny model of schizophrenia
It has been observed that certain developmental environmental risk factors for schizophrenia when modeled in rodents alter the trajectory of dopaminergic development, leading to persistent behavioural changes in adults. This has recently been articulated as the "dopamine ontogeny hypothesis of schizophrenia". To test one aspect of this hypothesis, namely that transient dopaminergic effects duri...
متن کاملThe Response of Ventral Tegmental Area Dopaminergic Neurons to Bupropion: Excitation or Inhibition?
Introduction: Antidepressants can modulate brain monoamines by acting on pre-synaptic and postsynaptic receptors. Autoreceptors can reduce the monoamines effect on the somatodendritic or pre-synaptic regions despite its postsynaptic counter effects. The direct effect of some antidepressants is related to its temporal and spatial bioavailability in the vicinity of these receptors (still a matter...
